Does this give 50 mana on kill also, or just for stamina? The items that listed to raise stamina did not raise my mana, so I wondered if this would be the same?

Yes. This restores mana as well. The ones that raise stamina will also raise mana. It may not show up until in battle, but it does. 200 is the mana/stamina cap, so if you use the great bear sigil you cannot increase it further.
